Decitabine therapy during Blast Crisis of CML

Categories:

Hello,

I have posted on this forum a few times regarding my 52yo mother who is currently undergoing treatment for the Blast Crisis of CML. She is currently taking 50mg Dasatinib twice a day(BD). We were previously on 50mg Dasatinib once a day(OD) but her counts have not really been reducing as quickly anymore and the doctor has increased dosage to BD, along with 100mg Cytosar for 3 days. After 3 days on cytosar, and 54 days on Dasatinib, her WBC counts are still ~15000. They were 60,000 last week.

Now we are being advised to take Decitabine along with Dasatinib after 10 days. Are there folks on here who have experienced this line of treatment in Blast crisis? How effective is the decitabine therapy?

Regards,
Sid

Hi Sid,

I found a presentation that mentions when decitabine is used with inecalcitol, it seems to provide better CML stem cell control.  Here's the source: https://www.google.com/url?sa=t&rct=j&q=&esrc=s&source=web&cd=9&cad=rja&...

But then I found that a study of this concept was stopped due to futility:  https://www.hybrigenics.com/contents/inecalcitol-2/acute-myeloid-leukemia

Also, asciminib is thought to be more effective when paired with a TKI.  Here's a link: https://www.ncbi.nlm.nih.gov/pmc/articles/PMC5862588/, and another:  https://clinicaltrials.gov/ct2/show/NCT02081378

These therapies haven't had much testing in humans yet, so it's unknown if the in vitro and mouse models will be replicated in humans.

I don't know if any of this information will help, but it might be worth talking to her doctor about it.

Hoping for the best, Kirk
