E450K Mutation

Hi all.

Are any of you familiar with the E450K mutation? My doctor tells me it is known for causing other mutations, but that's all the info I have.

I was on Imatinib for a year then it stopped working (which was when the mutation test was taken), then Dasatinib for 6 months but had to stop it as one of my lungs filled up with fluid - it was working well too but the side affects were bad, and now on Nilotinib which I'm handling ok so far at least.

The mutation test took about 6 months to come back so only finding about this now. I tried Google but there isn't much information out there at all.

I would appreciate any info anyone has.

From a cursory look it appears that some mutations transform to T315I.  So maybe ponatinib or asciminib would be something to consider.

The E450K mutation can come and go, but it is NOT a precursor to other mutations.  It is a mutation which can be transient, which is not unusual.  I would suggest a re-do of the mutation test.  E450K is not considered to be a difficult mutation to overcome if it does persist, which it may not.  But if it persists then Ponatinib may be the preferred TKI.
